Physiologically based pharmacokinetic (PBPK) modeling provides quantitative descriptions of chemical fate in the body through absorption, distribution, metabolism, and excretion processes. PBPK models can be used in myriad risk assessment applications by allowing toxicology data to be translated across species, between exposure routes, and among potentially exposed or susceptible subpopulations. Over the past several years, ScitoVation has created a flexible, open-source modeling platform called Population Life-course Exposure to Health Effects Model (PLETHEM). PLETHEM combines high-throughput exposure prediction programs and rapid physiologically based pharmacokinetic modeling (PBPK) into a single framework, making these tools more accessible to more practitioners. The development of PLETHEM has drawn on the advanced modeling technologies pioneered by ScitoVation scientists under the continuing support of the ACC LRI. PLETHEM provides a previously unavailable capability for rapidly predicting chemical dosimetry to support risk assessments ranging from high-throughput screening to TSCA risk evaluations, as evidenced by the fact that the EPA Office of Pesticides utilized PLETHEM as a user-friendly, open-source platform for their evaluations of PBPK models submitted in support of pesticide risk evaluations.
