To move toward replacement of animal models in chemical risk assessment, the field must continue to develop fit-for- purpose in vitro assays that examine cellular pathway responses in models that accurately reflect the relevant biology of the in vivo system where effects occur.

To determine toxicological effects, these in vitro assays must be:

  • Consistent with the known biology of the relevant toxicity pathway
  • Capable of recapitulating the phenotype of interest with measures of adversity (i.e., estrogen-induced proliferation, nuclear receptor activation, etc.)
  • Able to produce responses at chemical concentrations that are consistent with in vivo results
